Eurostars Project E!11034 CRYOSPHERE: 3D co-cultures of human PC and NPC cells from same liver: Prediction of drug-induced liver injury

Reference number
Coordinator Karolinska Institutet - Department of Physiology and Pharmacology
Funding from Vinnova SEK 2 000 000
Project duration March 2017 - February 2019
Status Completed
Venture Eurostars

Purpose and goal

The Project was very successful. We created genotypically and phenotypically characterized Spheroid PHH and NPC biobanks and made selection of the best donors. We developed and validation of spheroid co-culture method of cryopreserved PHH and NPC from the same donors as a model and got successful results. The spheroids were phenotypically characterized with respect to genes expressed, protperties and stability. We could successfuly develop a novel in vitro model for NASH.

Expected results and effects

The spheroid systems developed will be of very high commercial interest for development of novel drugs against NASH. Also the characterization and validation of donors for the spheroid formaiton will be of utmost importance for commersialization of the spheorid system by KaLy cell.

Planned approach and implementation

We had continuous contacts with KaLy Cell and two F2F meetings as well as mail and telephone conferences. The screening of cells occurred in Strasbourg while the formation of spheroids and examination of mechanisms and conditions for fibro formation occurred at Karolinska Institutet. Overall, the cooperation worked very well.
