Important results from the project
This project aims to decipher the mechanism underlying the involvement of peroxynitrite in opioid tolerance setting and if the working theory of the endogenous morphine would be possible in case of exogenous administrated opiates. A full plan to detect the peroxynitrite in the important brain regions involved in pain and its marker.
Expected long term effects
The Karolinska institute has a core facility and animal houses and all the equipment needed in this project. Animal experiments and cell culture are important models. Therefore, everything is well prepared for the success of the technical procedure. As for the scientific competence, the host group collaborating with other expertise in the institute and other universities in Sweden and we made sure that the required scientific advice will be available.
Approach and implementation
The project description and planning is based on molecular biology and animal experimentation. The animals will be divided into drug treated and controls. The expression of associated genes and proteins will be assessed by quantitative polymerase reaction and protemics (Western blot). Comparison between the groups by ANOVA. The peroxinitrite will be measured by ELISA. The markers for peroxynitrite as nitrotyrosine will be assessed also by Western blotting.
