Purpose and goal

The aim of the project was to test if magnetic separation of biotherapeutic antibodies from cell culture is an alternative to classical column chromatography. To show this, the experiments must be performed on a larger pilot scale. The goal of the project, with a positive outcome in pilot scale, would allow for further development of equipment for industrial process scale.

Expected results and effects

The project goals have been achieved. We have shown that magnetic separation of antibodies produced in cells works on a pilot scale, and this provides attractive benefits over classic column chromatography strategies. Two bioprocess companies have expressed their spontaneous interest in the LOABeads System for magnetic purification of biologics. The next goal is to build a product prototype and test at an industrial site.

Planned approach and implementation

The project has been a collaboration between LOAB and KTH, where LOAB has developed and further developed a magnetic proof-of-concept separation prototype for pilot scale. KTH has grown antibody producing cells and performed initial magnetic bead purification experiments at small lab scale. Later, the two subprojects have been intertwined and moved on to purifications in larger pilot scale. The two parties have worked closely throughout the project, with much contact by phone, mail, Skype and physical presence at both places.
