Purpose and goal
The aim of STAMINA is to develop a new class of GRK2-biased β2-agonists with sustained, non-desensitizing muscle-building effects against muscle-wasting diseases — primarily FSHD, but also DMD, SMA and sarcopenia. Previous β2-agonists have shown muscle-building effects in patients but have lost efficacy over time; STAMINA addresses this. The goal is a complete preclinical basis for a drug candidate, ready for clinical trials and future partnerships with the pharmaceutical industry.
Expected effects and result
STAMINA is expected to deliver a complete preclinical basis for a drug candidate (CD package) against muscle wasting diseases, validated in patient-derived disease models and ready to enable proposal for clinical trials. The project strengthens Atrogi´s platform and intellectual property rights in FSHD, DMD, SMA and sarcopenia, and paves the way for new treatments for patients without approved treatment, a future partnership with the pharmaceutical industry and strengthened Swedish life science.
Planned approach and implementation
STAMINA is being implemented by six partners in Sweden, the Netherlands and Latvia over a period of 36 months. Novel β2-agonists are designed and synthesized, screened in vitro and in human muscle cells (Karolinska) and tested in disease-specific models — patient-derived 3D-FSHD-modeller (LUMC), SMA models (SU) and in vivo — as well as mechanistically.
